目的:从加味茵芍散对肝纤维化家兔转化生长因子-β1(TGF-β1)、丝/苏氨酸激酶2(Smad2)、丝/苏氨酸激酶4(Smad4)mRNA表达的影响探讨该方对肝纤维化的可能作用机制。方法:按0.10mL/kg的用量腹腔注射CCl4诱导复制家兔肝纤维化模型,50只家兔随机分成模型组,秋水仙碱组,加味茵芍散高、中、低剂量组5组,每组10只,通过肝脏病理取材确定肝纤维化病变,同时实时定量逆转录一聚合酶链反应法检测TGF-β1、Smad2 mRNA、Smad4 mRNA的表达。结果:与模型组相比,秋水仙碱组和加味茵芍散高、中、低剂量组家兔肝组织TGF-β1、Smad2 mRNA、Smad4 mRNA表达明显减弱(P<0.05);加味茵芍散中、高剂量组与秋水仙碱组相比TGF-β1、Smad2 mRNA、Smad4 mRNA的表达量显著降低(P<0.05)。结论:加味茵芍散能减轻纤维组织增生,可以下调TGF-β1、Smad2 mRNA、Smad4的表达。该方对CCl4诱导家兔肝纤维化模型具有一定保护作用、可以延缓肝纤维化的进展,其作用机制可能与调节TGF-β/Smad信号通路有关。
Objective: To investigate the effect of Jiawei Yinshao Powder on the expression of TGF-β1, Smad2 and Smad4 mRNA in liver fibrosis rabbits The side of the possible mechanism of liver fibrosis. Methods: CCl4 was intraperitoneally injected at a dose of 0.10 mL / kg to induce the model of hepatic fibrosis in rabbits. Fifty rabbits were randomly divided into 5 groups: model group, colchicine group, Jiawei YinShao Powder high, medium and low dose groups The rats in group A were divided into three groups according to their liver pathology. The pathological changes of hepatic fibrosis were confirmed by real-time RT-PCR. The expressions of TGF-β1, Smad2 mRNA and Smad4 mRNA were detected by real-time reverse transcription-polymerase chain reaction. Results: Compared with model group, the expressions of TGF-β1, Smad2 mRNA and Smad4 mRNA in hepatic tissue of colchicine group and Jiawei YinShao-San high, medium and low dose group were significantly decreased (P <0.05) The mRNA expression of TGF-β1, Smad2 and Smad4 in middle-dose and high-dose groups were significantly lower than those in colchicine group (P <0.05). Conclusion: Jiawei YinShao Powder can reduce the proliferation of fibrous tissue, and down-regulate the expression of TGF-β1, Smad2 mRNA and Smad4. This prescription can protect CCl4-induced hepatic fibrosis model and delay the progression of hepatic fibrosis, and its mechanism may be related to the regulation of TGF-β / Smad signaling pathway.
