目的 观察热疗和顺铂对放射治疗的增强作用,以探索一种临床更为有效的肿瘤治疗方法。方法 采用X线照射前或后的人卵巢癌细胞A2780S( 对顺铂敏感) 和A2780CP( 抗顺铂) 用顺铂和热疗处理,观察两者对亚致死剂量辐射损伤修复(SLDR) 的抑制作用。结果 热疗(40℃,1 小时) 或顺铂处理1 小时均不能抑制这一修复,但两者联合应用则对两个细胞系均有明显效果。SLDR 的受抑程度取决于处理的时机。对于抗顺铂的A2780CP,可观察到SLDR的抑制,而在敏感细胞A2780S,除此之外还能观察到放疗敏感性的提高。结论 单独应用作用有限的热疗和低浓度顺铂联合应用可明显增强放疗疗效。
Objective To observe the enhanced effect of hyperthermia and cisplatin on radiation therapy in order to explore a clinically more effective method of tumor treatment. METHODS: Human ovarian cancer cells A2780-S (sensitive to cisplatin) and A2780-CP (anti-cisplatin) were treated with X-ray before or after irradiation with cisplatin and hyperthermia to observe their effects on sublethal dose radiation damage repair ( Inhibition of SLDR). RESULTS: Hyperthermia (40°C, 1 hour) or cisplatin treatment for 1 hour did not inhibit the repair, but the combination of the two had significant effects on both cell lines. The degree of inhibition of SLDR depends on the timing of the treatment. For the anti-cisplatin A2780-CP, SLDR inhibition was observed, but in sensitive cells A2780-S, in addition to the sensitivity of radiotherapy can be observed. Conclusion The combination of hyperthermia with limited effect and low concentration of cisplatin alone can significantly enhance the efficacy of radiotherapy.
