目的探讨回转模拟微重力对人的血管内皮细胞形态、NO产生及一氧化氮合酶表达的影响。方法利用体外培养的人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVEC),采用回转器模拟微重力效应,在微重力条件下培养人脐静脉内皮细胞48h,同时设1g静止培养对照。倒置显微镜下观察细胞生长形态,透射电镜观察HUVEC超微结构。检测不同转速下细胞产生NO的量,及细胞中内皮型一氧化氮合酶(eNOS)、诱导型一氧化氮合酶(iNOS)的表达。结果微重力培养后,细胞变圆且重叠生长;回转组NO产生量高于1g对照组且与一定的回转速度呈正相关。回转组细胞eNOS表达较对照组增多且表达iNOS,对照组细胞未表达iNOS。结论微重力对体外人脐静脉内皮细胞的形态,NO产量及NOS的合成都有影响。
Objective To investigate the effects of gyro-simulated microgravity on human vascular endothelial cell morphology, NO production and nitric oxide synthase expression. Methods Human umbilical vein endothelial cells (HUVEC) cultured in vitro were used to simulate the effects of microgravity by gyroscope. Human umbilical vein endothelial cells (HUVECs) were cultured under microgravity conditions for 48 hours. At the same time, 1g static culture control was set. The morphology of HUVEC was observed under inverted microscope. The ultrastructure of HUVEC was observed by transmission electron microscopy. The amount of NO produced by cells at different rotational speed and the expression of eNOS and iNOS in cells were detected. Results After microgravity culture, the cells became round and overlapped. The amount of NO produced in swirling group was higher than that in 1 g control group and positively correlated with certain rotational speed. The expression of eNOS in the swollen group was higher than that in the control group, and the expression of iNOS was not observed in the control group. Conclusion Microgravity has effects on the morphology, NO production and NOS synthesis of human umbilical vein endothelial cells in vitro.
