目的:检测RhoA、Ezrin和CD44蛋白在喉鳞状细胞癌(LSCC)组织中的表达,探讨三者在LSCC浸润转移中的作用及其相关性。方法:应用免疫组织化学SABC法检测68例LSCC组织及35例喉正常鳞状上皮组织中RhoA、Ezrin和CD44蛋白的表达水平,并结合临床病理特征进行统计学分析。结果:RhoA、Ezrin和CD44蛋白在LSCC组织的阳性表达率分别为80.9%、82.4%和70.6%,在喉正常鳞状上皮组织中的阳性表达率分别为28.5%、25.7%和20.0%。2组间的3种蛋白表达比较差异均有统计学意义(P<0.05)。RhoA、Ezrin和CD44表达水平与LSCC患者的年龄、性别、吸烟与否、肿瘤部位无关(P>0.05),而与肿瘤的分化程度、浸润程度、淋巴结的转移、TNM分期密切相关(P<0.05)。LSCC组织中RhoA、Ezrin和CD44蛋白表达呈正相关。结论:RhoA、Ezrin和CD44蛋白在LSCC的浸润转移中均发挥重要作用,提示三者在该过程中起协同作用。
OBJECTIVE: To detect the expression of RhoA, Ezrin and CD44 in laryngeal squamous cell carcinoma (LSCC) tissues and to explore their roles in the invasion and metastasis of LSCC. Methods: The expressions of RhoA, Ezrin and CD44 in 68 cases of LSCC and 35 cases of laryngeal normal squamous epithelium were detected by immunohistochemical SABC method, and their clinical pathological characteristics were analyzed statistically. RESULTS: The positive rates of RhoA, Ezrin and CD44 in LSCC tissues were 80.9%, 82.4% and 70.6%, respectively. The positive rates of RhoA, Ezrin and CD44 in normal laryngeal tissues were 28.5%, 25.7% and 20.0%, respectively. There were significant differences in the expression of the three proteins between the two groups (P <0.05). The expression levels of RhoA, Ezrin and CD44 were not related to the age, sex, smoking or not of tumor and the location of tumor (P> 0.05), but closely correlated with the degree of differentiation, degree of invasion, lymph node metastasis and TNM stage ). The expressions of RhoA, Ezrin and CD44 in LSCC were positively correlated. CONCLUSION: RhoA, Ezrin and CD44 proteins play an important role in the invasion and metastasis of LSCC, suggesting that the three play a synergistic role in this process.
