目的通过研究环氧合酶(COX)-2反义寡核苷酸对乳腺癌细胞MDA-MB-231的增殖抑制作用并分析其抑制原理,探讨COX-2反义寡核苷酸对乳腺癌的作用意义。方法 MDA-MB-231细胞分为3组培养:对照组、正义寡核苷酸组及反义寡核苷酸组。MTT实验检测细胞的增殖情况;转染48 h后,Western blot检测COX-2蛋白的表达;流式细胞仪检测细胞周期的变化。结果与对照组、正义寡核苷酸组比较,COX-2反义寡核苷酸转染的细胞增殖受到明显抑制;蛋白表达明显下调;G0/G1期细胞比例明显增多,S期细胞所占比例显著降低(P均<0.05)。结论COX-2反义寡核苷酸可以抑制乳腺癌细胞MDA-MB-231细胞的增殖;下调COX-2蛋白的表达;并调节细胞周期的分布。
OBJECTIVE: To investigate the inhibitory effect of cyclooxygenase (COX) -2 antisense oligonucleotide on the proliferation of breast cancer cell line MDA-MB-231 and to analyze its inhibitory mechanism. Conclusion COX-2 antisense oligonucleotide can inhibit the proliferation of breast cancer cell MDA-MB-231 cells, down-regulate the expression of COX-2 protein and regulate the cell cycle distribution.
