目的测定培养的人眼小梁细胞糖胺多糖(glycosaminoglycan,GAG)的构成比。方法在体外培养从无眼病尸体眼取得的小梁细胞(trabecularmeshworkcel,TMC),掺入氚标葡萄糖胺,再用生物化学方法测定细胞层及培养液中的糖胺多糖,包括依次酶降解、醋酸纤维电泳及液体闪烁放射活性测定等。结果共获得5种糖胺多糖,其构成比是透明质酸30.5%、硫酸软骨素21.7%、硫酸皮质素14.9%、硫酸类肝素19.2%、硫酸角质素5.4%及其他物质8.3%。结论此结果与直接从人眼小梁细胞组织所测得的糖胺多糖的构成比颇为接近,可为进一步研究各种因素对人眼小梁细胞糖胺多糖代谢的影响及开角青光眼的发病学提供基础。
Objective To determine the composition ratio of glycosaminoglycan (GAG) in cultured human trabecular meshwork cells. Methods Trabecular mesothelioma (TMC) was obtained from cornea without cornea eye in vitro and mixed with tritiated glucosamine. The glycosaminoglycans in the cell layer and culture medium were determined by biochemical methods, including enzymatic degradation followed by acetic acid Fiber electrophoresis and liquid scintillation radioactivity determination. As a result, five glycosaminoglycans were obtained, the constituent ratio of which was 30.5% of hyaluronic acid, 21.7% of chondroitin sulfate, 14.9% of cortisone sulfate, 19.2% of heparan sulfate, and 5 keratan sulfate. 4% and other substances 8.3%. Conclusion This result is close to the composition ratio of glycosaminoglycans directly measured from human eye trabecular meshwork cells, which can be used to further study the effect of various factors on the metabolism of glycosaminoglycan in human trabecular meshwork cells and the effect of open-angle glaucoma Pathology to provide the foundation.
