目的:研究抗角蛋白自身抗体(AKautoAb)对白细胞介素1β(IL-1β)诱发角朊细胞增殖的影响.方法:采用角朊细胞无血清培养方法,以3H-TdR标示角朊细胞增殖程度,检测IL-1β刺激角朊细胞及AKau-toAb处理角朊细胞的增殖代谢.结果:1600U/ml及以上浓度IL-1β对角朊细胞增殖有明显促进作用(P<0.01),正常血清浓度AKautoAb对IL-1β引起的角朊细胞增殖有抑制作用(P<0.01).结论:AKautoAb抑制IL-1β引起的角朊细胞增殖,提示皮肤炎症反应中,角朊细胞内沉积的AKautoAb对角朊细胞的过度增殖有调节作用.
Objective: To investigate the effect of anti-keratin autoantibodies (AKautoAb) on the proliferation of keratinocytes induced by interleukin-1β (IL-1β). Methods: Proliferation of keratinocytes was detected by 3H-TdR and the proliferation of keratinocytes stimulated by IL-1β and AKau-toAb were detected by serum-free culture of keratinocytes. Results: The proliferation of keratinocytes was significantly enhanced at a concentration of 1600 U / ml and above (P <0.01). The normal serum concentration of AKautoAb could inhibit the proliferation of keratinocytes induced by IL-1β (P <0.01). 01). CONCLUSION: AKautoAb inhibits the proliferation of keratinocytes induced by IL-1β, suggesting that AKautoAb deposited in keratinocytes may play a regulatory role in the hyperproliferation of keratinocytes in the skin inflammatory response.
