目的:观察黄芩、柴胡提取物对CVB3m小鼠心肌炎凋亡及相关基因Bc l-2/Bax表达的影响。方法:采用腹腔注射CVB3m病毒诱导Balb/c小鼠心肌炎模型,采用TUNEL检测心肌组织细胞凋亡,免疫组化法检测心肌组织Bc l-2、Bax蛋白表达,逆转录聚合酶链式反应(RT-PCR)技术检测小鼠Bc l-2/BaxmRNA表达。结果:与模型组比较,黄芩、柴胡组心肌组织的凋亡细胞指数均明显增高;两组心肌组织中的Bc l-2表达明显下降,而黄芩组下降尤为明显;两组Bax的表达与模型组相比差异不显著。在病毒感染的3、5、7天的病理积分与Bc l-2蛋白阳性表达强度呈明显正相关。结论:黄芩、柴胡在病毒性心肌炎的急性期具有减轻心肌的病理损伤作用,其机制可能与其促进受染细胞的凋亡有关,而黄芩的作用表现尤为明显。
Objective: To observe the effects of Radix Scutellariae and Radix Bupleuri extract on the apoptosis of myocarditis and the expression of Bcl-2 / Bax in CVB3m mice. Methods: The model of Balb / c mouse myocarditis was induced by intraperitoneal injection of CVB3m virus. The apoptosis of myocardial cells was detected by TUNEL. The expressions of Bcl-2 and Bax in myocardium were detected by immunohistochemistry. The expression of Bcl- -PCR) technique was used to detect the Bcl-2 / Bax mRNA expression in mice. Results: Compared with the model group, the apoptotic cells index of Scutellaria baicalensis and Bupleurum group were significantly increased; the expression of Bcl-2 in both groups significantly decreased, while the Scutellaria group decreased significantly; the expression of Bax The difference between the model group was not significant. Pathological scores at 3, 5 and 7 days after virus infection were positively correlated with the positive expression of Bcl-2 protein. Conclusion: Scutellaria baicalensis and Radix Bupleuri can attenuate the pathological damage of myocardium in the acute phase of viral myocarditis. The mechanism may be related to the promotion of apoptosis in infected cells, and the effect of Scutellaria baicalensis is more obvious.
