目的:观察加味桃核承气汤对四氯化碳(CCl4)肝纤维化大鼠α-平滑肌肌动蛋白(α-SMA)的影响。方法:用CCl4诱导大鼠肝纤维化模型,随机分为正常组、模型组,加味桃核承气汤高、中、低剂量组和秋水仙碱组;采用灌胃给药,疗程8周;用苦味酸-天狼红胶原染色方法进行半定量肝纤维化计分(SSS),观察肝纤维化程度的变化;用免疫组化技术半定量检测肝组织α-SMA的表达变化。结果:模型组大鼠SSS水平显著高于正常组,经加味桃核承气汤治疗后肝纤维化SSS水平明显下降;正常组、模型组,加味桃核承气汤高、中、低剂量组和秋水仙碱组肝组织α-SMA的蛋白表达水平分别为1.68±0.51、8.74±2.40、4.52±1.34、5.47±2.01、6.50±1.67、7.05±1.53。各治疗组α-SMA的蛋白表达水平显著下降。结论:加味桃核承气汤对CCl4诱导的肝纤维化有显著的治疗作用,其作用机制与抑制HSC的活化有关。
OBJECTIVE: To observe the effect of Jiawei Taohe Chengqi Decoction on carbon tetrachloride (CCl4) fibrosis in rats with α-smooth muscle actin (α-SMA). Methods: Rat models of hepatic fibrosis induced by CCl4 were randomly divided into normal group and model group, Jiawei Taohe Decoction decoction high, middle and low dose group and colchicine group. The rats were given intragastric administration for 8 weeks. Semi-quantitative hepatic fibrosis scoring (SSS) was performed with picric acid-Sirius red collagen staining method to observe the changes of hepatic fibrosis. The expression of α-SMA in hepatic tissue was detected semiquantitatively by immunohistochemical technique. Results: The level of SSS in rats of model group was significantly higher than that of normal group. The level of SSS of hepatic fibrosis after treatment with Modified Taohe Chengqi Decoction was significantly decreased. Normal group, model group, Jiawei Taohe Chengqi Decoction high, middle and low dose group The protein expression levels of α-SMA in the liver and colchicine groups were 1.68±0.51, 8.74±2.40, 4.52±1.34, 5.47±2.01, 6.50±1.67, and 7.05±1.53, respectively. The protein expression levels of α-SMA were significantly decreased in each treatment group. CONCLUSION: Modified Taohe Chengqi Decoction has a significant therapeutic effect on CCl4-induced liver fibrosis, and its mechanism of action is related to the inhibition of HSC activation.
