[目的]利用分子标记SRAP技术,构建22个甘薯品种的DNA指纹图谱并进行遗传多样性分析。[方法]从49对SRAP引物中筛选了11对引物进行PCR扩增。[结果]共扩增出98条带,多态性条带74条,多态性比率为75.5%,平均每对引物扩增8.91条带。利用引物Me2-em3和Me3-em7,构建了22个甘薯品种的指纹图谱。聚类结果显示,在阈值为15.5时可将22个甘薯品种被分为7类。[结论]地理来源相同的甘薯品种和具有同一亲本的甘薯品种聚在了一起。
[Objective] The research aimed to construct DNA fingerprinting of 22 sweet potato cultivars and analyze the genetic diversity by molecular marker SRAP technology. [Method] Eleven pairs of primers were screened from 49 pairs of SRAP primers for PCR amplification. [Result] A total of 98 bands and 74 polymorphic bands were amplified, with a polymorphism ratio of 75.5%. An average of 8.91 bands per pair of primers were amplified. Fingerprints of 22 sweet potato cultivars were constructed using primers Me2-em3 and Me3-em7. Cluster analysis showed that 22 sweet potato cultivars could be divided into 7 groups at a threshold of 15.5. [Conclusion] Sweet potato varieties with the same geographical origin and sweet potato varieties with the same parents were clustered together.
