目的:探讨南蛇藤提取物对人食管鳞癌TE-8细胞凋亡的影响,并对其机制进行初步探讨。方法:MTT法检测不同质量浓度的南蛇藤提取物作用于TE-8细胞24、48h后细胞的增殖情况。南蛇藤提取物作用TE-8细胞24h后,Annexin V-FITC/PI双染法检测细胞凋亡情况;JC-1试剂盒检测线粒体膜电位的损伤;TUNEL法检测DNA损伤情况;Western blot法检测丝裂原活化的蛋白激酶(MAPK)信号通路相关蛋白的表达情况。结果:南蛇藤提取物能明显抑制人食管鳞癌TE-8细胞的增殖,呈时间及浓度依赖;南蛇藤提取物能促进细胞凋亡,降低线粒体膜电位,诱导细胞内DNA损伤,阻断MAPK信号通路的表达,呈浓度依赖。结论:南蛇藤提取物能够抑制TE-8细胞的增殖,促进其凋亡,其机制可能与南蛇藤提取物抑制MAPK信号通路的表达有关。
Objective: To investigate the effects of Celastrus orbiculatus extract on the apoptosis of human esophageal squamous cell carcinoma TE-8 cells, and to explore its mechanism. Methods: The proliferation of TE-8 cells treated with different concentrations of Celastrus orbiculatus extract for 24,48 hours was detected by MTT assay. The apoptosis of mitochondrial membrane potential was detected by JC-1 kit, the DNA damage was detected by TUNEL method and the Western blot method was used to detect the apoptosis of TE-8 cells after 24 hours. The expression of mitogen-activated protein kinase (MAPK) signaling pathway-related proteins was detected. Results: The extract of Celastrus orbiculatus could significantly inhibit the proliferation of human esophageal squamous cell carcinoma TE-8 cells in a time and concentration-dependent manner. Celastrus orbiculatus extract could promote apoptosis, reduce mitochondrial membrane potential and induce DNA damage in the cells Disruption of MAPK signaling pathway in a concentration-dependent manner. CONCLUSION: The extract of Celastrus orbiculatus can inhibit the proliferation and promote the apoptosis of TE-8 cells. The mechanism may be related to the inhibition of MAPK signaling pathway by Celastrus ornithus extract.
