目的探讨桑色素对人视网膜母细胞瘤细胞增生抑制及诱导凋亡的作用。设计实验研究。研究对象人视网膜母细胞瘤细胞Y79细胞。方法不同浓度(0、50、100、150、250μM)桑色素对人视网膜母细胞瘤细胞Y79细胞作用24、48、72小时后,CCK8法检测肿瘤细胞生存率的变化;细胞周期检查法检测肿瘤细胞周期的变化;用TUNEL法检测肿瘤细胞凋亡的变化。主要指标细胞生存率、细胞周期分布、细胞凋亡率。结果桑色素浓度为150、250μM,与桑色素浓度为0、50、100μM作用于Y79细胞相比,检测时间分别在48、72小时时的细胞生存率显著下降(P<0.05),细胞周期G0-G1期显著增多(P<0.05),S期显著下降(P<0.05),细胞凋亡显著增多(P<0.05)。较高浓度的桑色素,随着作用时间的延长,引起Y79细胞生存率逐渐下降,细胞凋亡率逐渐增多,具有时间依赖性和剂量依赖性。结论桑色素能够抑制视网膜母细胞瘤增生并促进其凋亡,提示桑色素有望成为治疗视网膜母细胞瘤的有效药物。
Objective To investigate the effect of morin on the proliferation and apoptosis of human retinoblastoma cells. Design experiment research. Study target human retinoblastoma cells Y79 cells. Methods Morin in different concentrations (0,50,100,150,250μM) of human retinoblastoma cells Y79 cells 24,48,72 hours after the detection of CCK8 tumor cell survival rate changes; cell cycle detection of tumor Cell cycle changes; TUNEL assay of apoptosis in tumor cells. The main indicators of cell survival rate, cell cycle distribution, apoptosis rate. Results Compared with Y79 cells treated with morin at 0, 50 and 100μM, the cell cycle of G0 was significantly lower at 48 and 72 hours (P <0.05) -G1 phase (P <0.05), S phase decreased significantly (P <0.05), apoptosis increased significantly (P <0.05). Higher concentration of morin, with the extension of time, caused the Y79 cell survival rate decreased, the rate of apoptosis increased gradually, with a time-dependent and dose-dependent. Conclusion Morin can inhibit retinoblastoma proliferation and promote apoptosis, suggesting that morin is expected to be an effective drug for the treatment of retinoblastoma.
