目的:观察反式白藜芦醇对自发性高血压大鼠心肌肥大及丝裂原活化蛋白激酶表达的影响。方法:21只SHR大鼠随机分为模型组、白藜芦醇高(45mg/kg)、低(15mg/kg)剂量组,另7只同龄WKY大鼠作为正常对照组。30天实验后,分别采用尾套法、称重与彩色多普勒超声法、定量PCR法检测各组大鼠血压、左心室肥大指标、以及心肌组织JNK mRNA和p38MAPK mRNA的表达。结果:反式白藜芦醇(45mg/kg)能显著降低SHR血压、左心室PWT、IVST及心肌肥厚指数,减少心肌组织中JNK、p38MAPKmRNA表达。结论:反式白藜芦醇能有效防治SHR大鼠心肌肥大,其作用与调控p38MAPK和JNK信号转导相关。
AIM: To observe the effects of trans-resveratrol on cardiac hypertrophy and mitogen-activated protein kinase in spontaneously hypertensive rats. Methods: Twenty-one SHR rats were randomly divided into model group, high dose of resveratrol (45mg / kg), low dose (15mg / kg) and another 7 WKY rats of same age as normal control group. After 30 days of experiment, tail cuff method, weighing and color Doppler ultrasound were used to detect the blood pressure, left ventricular hypertrophy index and the expression of JNK mRNA and p38MAPK mRNA in myocardium by quantitative PCR. RESULTS: Trans resveratrol (45 mg / kg) significantly reduced SHR blood pressure, left ventricular PWT, IVST and cardiac hypertrophy index, and decreased the expression of JNK and p38 MAPK mRNA in myocardium. CONCLUSION: Trans-resveratrol can effectively prevent cardiac hypertrophy in SHR rats and its effect is related to the regulation of p38 MAPK and JNK signal transduction.
