目的观察人白细胞干扰素(IFN-α)对子宫内膜腺癌细胞系(JEC)细胞生长和细胞周期的影响。方法用不同浓度:2000U/ml、500U/ml、100U/ml的IFN-α与JEC共同培养8d,观察JEC细胞生长曲线、细胞凝集素受体表达变化,并用流式细胞仪分析癌系细胞周期。结果和结论IFN-α对JEC细胞的增殖有抑制作用,实验浓度越大,抑制作用越明明,但不影响凝集素受体的表达;IFN-α可使JEC细胞G1/G0阻滞,S期积累和G2/M期减少,前者与IFN-α浓度呈正相关,而后者则为负相关。
Objective To observe the effect of human leukocyte interferon (IFN-α) on the cell growth and cell cycle of endometrial adenocarcinoma cell line (JEC). Methods The JEC cell growth curve and the expression of lectin receptor were observed after co-cultured with JEC for different time with different concentrations of 2000U / ml, 500U / ml and 100U / ml IFN-α for 8 days. The cell cycle of cancer cells was analyzed by flow cytometry . RESULTS AND CONCLUSION IFN-α could inhibit the proliferation of JEC cells. The greater the experimental concentration, the more obvious the inhibitory effect was, but did not affect the expression of lectin receptor. IFN-α could block the arrest of G1 / G0 in JEC cells, Accumulation and G2 / M phase decreased, the former was positively correlated with the concentration of IFN-α, while the latter was negatively correlated.
