目的建立一种用于测定非洲木犀榄叶中木犀草苷、橄榄苦苷含量的HPLC方法。方法采用Thermo Syncronis C18柱(4.6 mm×250 mm,5μm),以乙腈-0.1%磷酸水为流动相,梯度洗脱,流速1.0ml·min-1,柱温30℃,检测波长为240 nm。结果木犀草苷、橄榄苦苷分别在13.95~558μg·ml-1(R2=0.9999)、24.5~980μg·ml-1(R2=0.9993)范围内线性关系良好,平均回收率(n=6)分别为101.25%(RSD=1.05%)、99.29%(RSD=1.98%)。结论建立的方法操作简便、结果准确,具有良好的重复性和稳定性,可用于非洲木犀榄叶中木犀草苷和橄榄苦苷的鉴定及质量评价。
OBJECTIVE To establish a HPLC method for the determination of luteolin and oleuropein in African rosewood leaves. The method was performed on a Thermo Syncronis C18 column (4.6 mm × 250 mm, 5 μm) using acetonitrile-0.1% phosphoric acid as mobile phase with a gradient of 1.0 ml · min-1 and a column temperature of 30 ° C. The detection wavelength was 240 nm. Results The results showed that there was a good linear relationship between luteolin and oleuropein in the range of 13.95-558 μg · ml-1 (R2 = 0.9999) and 24.5-980 μg · ml-1 (R2 = 0.9993), respectively 101.25% (RSD = 1.05%), 99.29% (RSD = 1.98%). Conclusion The established method is simple, accurate, reproducible and stable. It can be used to identify and evaluate the quality of luteolin and oleuropein in P. africanum.
