目的:观察非诺贝特对溶血卵磷脂(LPC)诱导的内皮细胞X连锁凋亡抑制蛋白(XIAP)基因表达的影响。方法:体外培养人脐静脉内皮细胞,分为:①正常对照组、②LPC组、③非诺贝特低浓度(10μmol/L)组、④非诺贝特中浓度(50μmol/L)组及⑤非诺贝特高浓度(100μmol/L)组。采用实时定量PCR及流式细胞仪分别观测LPC对内皮细胞XIAP mRNA及蛋白表达的影响,及非诺贝特干预后的变化。结果:与正常对照组比较,LPC使XIAP mRNA及蛋白表达减弱。非诺贝特干预后内皮细胞XIAP mRNA及蛋白表达增强。结论:非诺贝特可通过促进XIAP基因表达干预LPC对内皮细胞凋亡的影响。
Objective: To observe the effects of fenofibrate on the expression of X-linked inhibitor of apoptosis protein (XIAP) gene induced by lysophosphatidylcholine (LPC) in endothelial cells. Methods: Human umbilical vein endothelial cells were cultured in vitro and divided into three groups: ① normal control group, ② LPC group, ③ low concentration of fenofibrate (10 μmol / L), ④ moderate concentration of fenofibrate (50 μmol / L) Fenofibrate high concentration (100μmol / L) group. Real-time quantitative PCR and flow cytometry were used to observe the effect of LPC on the expression of XIAP mRNA and protein in endothelial cells and the changes after fenofibrate intervention. Results: Compared with the normal control group, LPC reduced the expression of XIAP mRNA and protein. Increased expression of XIAP mRNA and protein in endothelial cells after fenofibrate intervention. Conclusion: Fenofibrate interferes with the effect of LPC on the apoptosis of endothelial cells by promoting the expression of XIAP.
