目的:探索建立肾阳虚肺纤维化大鼠模型。方法:SD大鼠30只,随机分为生理盐水组(NS)、肺纤维化组(BLM)、肾阳虚纤维化组(RYA),每组10只,于一次性气管内滴入博莱霉素后,每日灌胃腺嘌呤,至第29天早上取标本处死大鼠,通过四诊表现、血肌酐(BUN)、尿素氮(Cr)、皮质醇(Cor)和肺肾病理改变及运用组织芯片结合胶原染色图象分析行肺胶原定量,鉴定肾阳虚肺纤维化大鼠模型。结果:肾阳虚肺纤维化组大鼠大体表现及肾病理改变与肾阳虚动物模型相似,肺病理改变与肺纤维化组相似,但肺胶原沉积程度(0.5413±0.2104)较肺纤维化组(0.9578±0.3756)轻(P<0.05),BUN、Cr(8.4872+2.8025和835.7333+377.2046)皆明显高于肺纤维化组(5.7863+1.3817和356.5400+300.5365)(P<0.05),Cor(14.1700+1.8999)则明显低于生理盐水组(20.4889+7.2648)和肺纤维化组(24.5040+9.4264)(P<0.05)。结论:肾阳虚肺纤维化大鼠同时具备了肾阳虚和肺纤维化的特征,说明模型新创成功,可作为进一步研究肾阳虚肺纤维化及温阳中药防治肺纤维化的动物模型。
Objective: To explore the establishment of rat model of kidney-yang deficiency pulmonary fibrosis. Methods: Thirty SD rats were randomly divided into normal saline group (NS), pulmonary fibrosis group (BLM) and kidney yang deficiency fibrosis group (RYA), with 10 rats in each group. After intragastric administration of adenine, the rats were sacrificed on the morning of the 29th day, and the pathological changes of serum creatinine (BUN), urea nitrogen (Cr), cortisol (Cor) Tissue microarray combined with collagen staining image analysis of pulmonary collagen quantitative determination of kidney yang deficiency pulmonary fibrosis rat model. Results: The general manifestation and renal pathological changes of rats with kidney-yang deficiency type of pulmonary fibrosis were similar to those of the animal model of kidney-yang deficiency. The pathological changes of lung were similar to those of pulmonary fibrosis group. However, the extent of pulmonary collagen deposition (0.5413 ± 0.2104) (0.9578 ± 0.3756), P <0.05, BUN and Cr (8.4872 + 2.8025 and 835.7333 + 377.2046) were significantly higher than those in pulmonary fibrosis group (5.7863 + 1.3817 and 356.5400 +300.5365, P <0.05) +1.8999) was significantly lower than the saline group (20.4889 + 7.2648) and pulmonary fibrosis group (24.5040 + 9.4264) (P <0.05). Conclusion: Kidney-Yang deficiency pulmonary fibrosis rats have the characteristics of both kidney-yang deficiency and pulmonary fibrosis, which indicates that the new successful model can be used as an animal model for further study of pulmonary fibrosis of kidney-yang deficiency and prevention and treatment of pulmonary fibrosis of warming Yang herbs .
