目的探讨四妙勇安汤对动脉粥样硬化家兔的干预效果及作用机理。方法36只日本雄性大耳白兔随机分为4组,分别为正常对照组、模型组、四妙勇安组及辛伐他汀组。通过高脂饲料、免疫损伤结合经股动脉球囊拉伤建立动脉粥样硬化家兔模型,四妙勇安组给予四妙勇安汤每次11.25g/kg生药量灌胃,每日2次;辛伐他汀组给予辛伐他汀5mg/kg灌胃,每日1次;正常对照组和模型组每日灌胃生理盐水20ml。于0、3、6、10周时测定各组家兔血超氧化物歧化酶(SOD)、丙二醛(MDA)、白细胞介素-1(IL-1)、肿瘤坏死因子α(TNF-α)、单核细胞趋化因子(MCP-1)的含量及主动脉细胞核因子-κB(NF-κB)的表达。结果与正常对照组比较,模型组各时间点MDA、IL-1、TNF-α、MCP-1水平显著增高(P<0.05或P<0.01),SOD与TNF-α之间存在一定相关性,四妙勇安组和辛伐他汀组在各时间点能够不同程度的调节MDA、IL-1、MCP-1、TNF-α、MCP-1水平及动脉壁NF-κB表达(P<0.05或P<0.01)。结论四妙勇安汤能够抑制动脉粥样硬化家兔炎症反应及氧化应激,增强兔动脉粥样硬化斑块的稳定性。
Objective To investigate the intervention effect and mechanism of Simiao Yong’an Decoction on atherosclerosis rabbits. Methods Thirty - six male Japanese white rabbits were randomly divided into 4 groups: normal control group, model group, Simiao Yongan group and Simvastatin group. Atherosclerosis rabbits were induced by high-fat diet, immunosuppression combined with femoral artery balloon injury. Simiao Yongan group was given Siqian Yongan Decoction 11.25g / kg crude drug for 2 times a day ; Simvastatin group given simvastatin 5mg / kg gavage once daily; normal control group and model group were given normal saline 20ml. The levels of serum superoxide dismutase (SOD), malondialdehyde (MDA), interleukin-1 (IL-1) and tumor necrosis factor-α (TNF- α), monocyte chemotactic factor (MCP-1) and the expression of nuclear factor-κB (NF-κB) in aorta. Results Compared with the normal control group, the levels of MDA, IL-1, TNF-α and MCP-1 in the model group at each time point were significantly increased (P <0.05 or P <0.01) The levels of MDA, IL-1, MCP-1, TNF-α, MCP-1 and the expression of NF-κB in arterial wall were significantly different in Simiao Yongan group and Simvastatin group at different time points (P <0.05 or P <0.01). Conclusion Simiao Yongan Decoction can inhibit inflammation and oxidative stress in rabbits with atherosclerosis and enhance the stability of atherosclerotic plaque in rabbits.
