目的 探讨+ GZ作用后,大鼠心肌组织内内皮素-1(ET-1)和内皮型一氧化氮合酶(eNOS)m RNA 表达变化。 方法 24只雄性Wistar 大鼠随机分成对照组、+ GZ重复暴露后30 m in、6 h 和24 h四组,每组6只。对照组大鼠G 值为+ 1 GZ, 实验组大鼠在动物离心机上经历了3次+ 10 GZ/3 m in(两次中间间隔30 m in)作用。分别于暴露后30 m in、6 h 和24 h 处死大鼠取心,提取总RNA,用半定量反转录聚合酶链反应(RT-PCR)检测方法检测大鼠心肌组织ET-1和eNOSm RNA 表达水平。 结果 + GZ暴露后30 m in 和6 h 大鼠心肌组织ET-1 m RNA 明显升高,eNOSm RNA 明显降低,但24 h 时均恢复正常。 结论 + GZ暴露可使大鼠心肌组织内ET-1和eNOSm RNA 表达发生改变,其在心肌损伤中可能起一定的作用,但这种变化是可逆的
Objective To investigate the expression of endothelin-1 (ET-1) and endothelial nitric oxide synthase (eNOS) m RNA in rat myocardium after + GZ treatment. Methods Twenty-four male Wistar rats were randomly divided into four groups: control group (n = 6) and control group (n = 6). The G value of the control group was + 1 GZ, and the rats in the experimental group underwent three +10 GZ / 3 mins (two intermediate intervals of 30 mins) on the animal centrifuge. The rats were sacrificed at 30 mins, 6 h and 24 h after exposure, respectively. The total RNA was extracted and the expression of ET-1 and eNOSm was detected by semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) RNA expression level. Results ET-1 m RNA in myocardial tissue of 30 min and 6 h after + GZ exposure was significantly increased, and eNOSm RNA was significantly decreased, but returned to normal after 24 h. Conclusion + GZ exposure can change the expression of ET-1 and eNOSmRNA in myocardial tissue of rats, which may play a role in myocardial injury, but this change is reversible
