目的:探讨二仙汤水提物对小鼠脾脏细胞和卵巢颗粒细胞作用的影响。方法:取小鼠脾脏和卵巢分别制备脾单个核细胞和卵巢颗粒细胞体外培养,加入二仙汤水提物,MTT法测细胞增殖情况;ELISA法测细胞上清液中IL-1、IL-5、IFN-γ以及雌二醇(E2)的浓度;荧光实时定量PCR检测雌激素受体(ERα、ERβ)及IL-1受体1(IL-1R1)的m RNA表达。结果:二仙汤对小鼠脾单个核细胞及卵巢颗粒细胞均具有较好的促增殖作用(P<0.01或P<0.05);可提高脾细胞培养上清中IL-1、IFN-γ及IL-5的含量(P<0.01或P<0.05),提高卵巢颗粒细胞上清中E2水平(P<0.05),使水提二仙汤组的Th1/Th2比值显著降低(P<0.05);并诱导脾细胞上ERα的m RNA表达(P<0.05)。结论:二仙汤水提物对小鼠免疫和内分泌功能均有较好的促进作用,可诱导Th1/Th2平衡向Th2偏移。
Objective: To investigate the effect of Erxian decoction on spleen cells and ovarian granulosa cells in mice. METHODS: Spleen mononuclear cells and ovarian granulosa cells were prepared from spleen and ovary of mice respectively. Water extract of Erxian decoction was added into the spleen and ovary to measure the proliferation of spleen and ovary. The proliferation of IL-1 and IL-5 , The concentrations of IFN-γ and estradiol (E2) were detected by real-time PCR. The m RNA expression of ERα, ERβ and IL-1R1 was detected by real-time quantitative PCR. Results: Erxian decoction could promote proliferation of spleen mononuclear cells and ovarian granulosa cells (P <0.01 or P <0.05), increase the contents of IL-1, IFN-γ, (P <0.01 or P <0.05), and increased the level of E2 in supernatant of ovarian granulosa cells (P <0.05), and significantly decreased the Th1 / Th2 ratio in water extract group (P <0.05). And induced the mRNA expression of ERα in spleen cells (P <0.05). Conclusion: The extract from Erxian decoction can promote the immune and endocrine function of mice, and induce the balance of Th1 / Th2 to Th2.
