目的改进制作角类药材组织切片的技术方法。方法采用滑走切片对角类药材进行常规切片取材,用质量分数为1%的番红溶液染色,用蒸馏水冲洗,用无水乙醇-冬青油(体积比为1∶1)溶液透明,最后用加拿大树胶封藏。结果采用此方法制片并在显微镜下观察发现,切片组织结构完整、细胞形态清晰、染色效果好,药材皮层组织、间质组织以及髓腔都清晰可见。结论该方法可为角类药材的真伪鉴别及教学质量的完善提供参考。
Objective To improve the technical method of making angle-sliced medicinal tissue sections. Methods Sliding sections were used for routine slicing of diagonal herbs, stained with Safranin at a mass fraction of 1%, rinsed with distilled water, transparent with a solution of absolute ethanol-oil of wintergreen (1: 1 by volume), and finally with Canada Gum sealed. Results The method of preparation and observation under the microscope showed that the tissue structure of the section was complete, the cell morphology was clear, the dyeing effect was good, and the cortex tissue, interstitial tissue and medullary cavity were all clearly seen. Conclusion This method can provide a reference for the authenticity and teaching quality of Coria medicinal herbs.
