[目的]探讨牛蒡苷(ARC)对弓形虫热休克蛋白70(T.g.HSP70)诱导弓形虫感染小鼠急性肝损伤的保护作用及其机制.[方法]将KM雌性小鼠随机分为正常组、模型组、ARC组、磺胺嘧啶钠阳性对照(PC)组.于弓形虫感染后第4d起给予药物治疗,连续5d.感染后第7d腹腔注射给予T.g.HSP70,次日末次给药4h后采集颈动脉血处死小鼠,取肝称质量,计算肝脏指数;采用比色法测定谷丙转氨酶(ALT)、谷草转氨酶(AST)水平、丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性;ELISA法测定细胞白细胞介素-1β(IL-1β)、肿瘤坏死因子α(TNF-α)水平;采用逆转录-聚合酶链式反应(RT-PCR)测定血小板活化因子(PAF-AH)mRNA的表达水平.[结果]与正常组比较,模型组ALT,AST水平和肝脏指数明显升高(P<0.05,P<0.01).与模型组比较,ARC组和PC组ALT,AST水平和MDA,IL-1β,TNF-α含量及PAF-AH mRNA表达水平均明显降低(P<0.05,P<0.01),SOD活性明显升高(P<0.05).ARC组与PC组上述各项指标间差异均无统计学意义(P>0.05).[结论]ARC对T.g.HSP70诱导弓形虫感染小鼠急性肝损伤具有保护作用,其机制可能与抗脂质过氧化及抑制炎症介质的产生有关.
[Objective] To investigate the protective effect and mechanism of arctiin (ARC) on acute liver injury induced by Toxoplasma gondii Toxoplasma gondii in mice induced by Toxoplasma gondii. [Methods] KM female mice were randomly divided into normal group, Group, ARC group and sodium sulfadiazine positive control group (PC) .After 4 days of infection with Toxoplasma gondii, the mice were treated with drug for 5 days continuously.The TgHSP70 was given intraperitoneally on the 7th day after infection, and the neck The mice were killed by arterial blood, the liver weight was taken and the liver index was calculated. The levels of ALT, AST, MDA and SOD were measured by colorimetric method. The levels of IL-1β and TNF-α were measured by ELISA. The levels of platelet-activating factor (PAF-AH) (P <0.05, P <0.01) .Compared with the model group, the levels of ALT and AST in the ARC group and the PC group were significantly higher than those in the normal group (P <0.05, P <0.01), and the activity of SOD increased significantly (P <0.05) .And the levels of MDA, IL-1β and TNF- (P> 0.05). [Conclusion] ARC has a protective effect on TgHSP70 induced acute liver injury in mice infected with Toxoplasma gondii, and its mechanism may be related to anti-lipid peroxidation and Inhibit the production of inflammatory mediators.
