目的:研究刺芒柄花素对大鼠血脂的调节作用和对肝脏雌激素受体表达的影响。方法:雌性SD成年大鼠40只,随机分为5组,即假手术组、模型组、尼尔雌醇组(阳性对照)、刺芒柄花素低剂量组、刺芒柄花素高剂量组,每组8只。假手术组打开腹腔后切除卵巢周围小块脂肪后关腹,后4组行双侧卵巢切除术,术后第5~10天每天1次进行阴道冲洗脱落细胞涂片,以了解去势是否彻底。确认去势成功后各组饲以高脂饮食,同时,假手术组和模型组灌胃生理盐水1mL.(100g)-1.d-1,尼尔雌醇组灌胃尼尔雌醇每周2.5mg·kg-1,刺芒柄花素低剂量组灌胃刺芒柄花素20mg·kg-1.d-1,刺芒柄花素高剂量组灌胃刺芒柄花素100mg·kg-1.d-1,连续8周。第8周后5d连续作阴道冲洗细胞涂片,第8周末断头处死大鼠,取血测血脂,取肝脏组织进行逆转录聚合本科链反应(RT-PCR),测定ERα、ERβ的相对表达量。结果:尼尔雌醇组和高剂量刺芒柄花素组血清TC、LDL-C明显降低,TG和HDL-C无明显改变;高剂量刺芒柄花素选择性上调去势大鼠肝脏ERβ表达。结论:刺芒柄花素能有效改善去势后高脂饮食大鼠血脂紊乱,这一作用可能是通过ERβ实现的。
OBJECTIVE: To study the regulatory effect of prothiranthon on serum lipids in rats and its effect on the expression of hepatic estrogen receptor. Methods: Forty female SD adult rats were randomly divided into five groups: sham operation group, model group, nilestriol group (positive control group), thiamarenate low dose group Group, 8 per group. Sham operation group after opening the abdominal cavity after the removal of small pieces of fat around the ovary and the abdomen, after 4 groups of bilateral ovariectomy, 5 to 10 days after surgery once a day for vaginal washing shedding cells smear in order to understand the castration is complete . After successful castration, rats in each group were fed with a high-fat diet, while sham-operated group and model group were given normal saline (1 mL) (100g) -1.d-1, nilestrol 2.5mg · kg-1, low-dose thymus-aconitine intragastric stomatalon 20mg · kg-1.d- -1.d-1 for 8 weeks. The rats were sacrificed at the end of the 8th week, and blood was collected for the detection of blood lipids. Reverse transcription-polymerase chain reaction (RT-PCR) was performed on the liver tissue to determine the relative expression of ERα and ERβ the amount. Results: The levels of TC and LDL-C in serum of nilestriol and high-intensity thymus tolunilat groups were significantly decreased, but TG and HDL-C did not change significantly. High doses of thorononetin selectively upregulated ERβ expression. Conclusion: Thimanchin can effectively improve the dyslipidemia in rats after castrated high-fat diet, and this effect may be achieved by ERβ.
