目的应用MTT法检测不同种类化疗药物对口腔癌细胞的体外抑制率,比较各类化疗药物抗肿瘤作用的差异,为口腔颌面部肿瘤临床治疗中化疗药物的筛查提供理论依据。方法应用MTT法检测环磷酰胺、氟尿嘧啶、平阳霉素、紫杉醇、顺铂等24种化疗药物对口腔癌细胞的体外抑制率,结果以珚x±s表示,应用多个独立样本非参数检验,通过比较不同化疗药物组间及组内对口腔癌细胞体外抑制率的不同,筛选对口腔癌细胞具有较高抑制作用的化疗药物。结果各种化疗药物对口腔鳞癌细胞均具有一定的抗肿瘤活性,五类化疗药物中植物类化疗药物对口腔鳞状细胞癌细胞的抑制率较其他种类化疗药物有显著性差异(P<0.05),其余各组内药物之间差异无统计学意义。结论各种化疗药物间体外肿瘤抑制率有差异,植物类抗肿瘤药物肿瘤抑制率优于其他化疗药物,其中紫杉醇的抗肿瘤活性最强,临床可选用作为首选药物,以期为患者带来更好的治疗效果。
Objective To detect the inhibitory rates of different kinds of chemotherapeutics on oral cancer cells in vitro by MTT assay and to compare the antitumor effects of various chemotherapeutic agents in order to provide a theoretical basis for the screening of chemotherapeutic drugs in the treatment of oral and maxillofacial tumors. Methods MTT assay was used to detect the in vitro inhibition rate of 24 chemotherapeutic drugs such as cyclophosphamide, fluorouracil, pingyangmycin, paclitaxel and cisplatin on oral cancer cells. The results were expressed as 珚 x ± s. Multiple independent samples were used for non-parametric tests. The chemotherapeutic agents with higher inhibitory effect on oral cancer cells were screened by comparing the inhibitory rates of oral cancer cells in vitro and in vivo between different chemotherapy groups. Results All kinds of chemotherapeutic drugs had certain antitumor activity on oral squamous cell carcinoma cells. The inhibitory rates of phytochemical drugs on oral squamous cell carcinoma cells in five kinds of chemotherapeutic drugs were significantly different from those of other chemotherapeutic drugs (P <0.05 ), The other groups within the drug difference was not statistically significant. Conclusions The inhibition rates of various chemotherapeutic drugs in vitro are different from each other. The tumor inhibition rate of plant antitumor drugs is superior to other chemotherapeutic drugs. Among them, paclitaxel has the strongest antitumor activity and is clinically available as the drug of choice in order to bring better results to patients Therapeutic effect.
