2月18日出版的Science报道了T24膀胱癌细胞系的C-ras~H转化基因和正常人体DNA中非转化的对应部分,其区别在于单个碱基的变化。这个碱基的改变导致C-ras~H编码的P21蛋白质中甘氨酸被缬氨酸所置换。现在Muschel等提出一个膀胱癌病人肿瘤的C-ras~H基因有一个相应的碱基变化的初步证据:即病人的正常组织含有与C-ras~H不能鉴别的基因。重要的是Muschel等人既未研究肿瘤组织的基因也未研究正常组织的基因能否转化NIH3T3细胞。
Science, published February 18, reports on the non-transformed counterparts in the C-ras-H transformed gene of T24 bladder cancer cell lines and in normal human DNA, with the difference being a single base change. This change of base results in the replacement of the glycine in the P21 protein encoded by C-ras-H by valine. Now Muschel et al. Propose a preliminary evidence of a corresponding base change in the C-ras-H gene in patients with bladder cancer: the patient’s normal tissues contain genes that are not identifiable to C-ras-H. Importantly, Muschel et al. Neither transformed the NIH3T3 cells nor studied the genes of tumor tissues nor the normal tissues.
