目的探讨不同浓度葛根素对大鼠肾小球系膜细胞增殖的影响及其作用机制。方法采用体外细胞培养技术,采用MTT检测不同浓度葛根素对大鼠肾小球系膜细胞增殖的量效和时效关系。分别采用RT-PCR和ELISA,检测不同浓度葛根素作用大鼠肾小球系膜细胞后,结缔组织生长因子mRNA和蛋白表达水平的变化。结果葛根素可抑制大鼠GMCs增殖(P<0.05),该增殖抑制作用呈浓度依赖模式。不同浓度葛根素作用于大鼠GMCs后,结缔组织生长因子mRNA和蛋白表达水平均随药物浓度增加而逐渐减弱(P<0.05)。结论葛根素对大鼠肾小球系膜细胞增殖有明显的抑制作用,其作用机制可能与下调结缔组织生长因子基因转录和表达水平有关。
Objective To investigate the effect of different concentrations of puerarin on the proliferation of glomerular mesangial cells in rats and its mechanism of action. Methods The in vitro cell culture technique was used to detect the dose-effect and time-effect relationship of different concentrations of puerarin on proliferation of glomerular mesangial cells in vitro. RT-PCR and ELISA were used to detect the changes of mRNA and protein levels of connective tissue growth factor after different concentrations of puerarin treated rat glomerular mesangial cells. RESULTS Puerarin inhibited the proliferation of rat GMCs (P<0.05). The inhibition of proliferation was concentration-dependent. After different concentrations of puerarin on rat GMCs, the expression of connective tissue growth factor mRNA and protein gradually decreased with increasing drug concentration (P<0.05). Conclusion Puerarin can significantly inhibit the proliferation of glomerular mesangial cells in rats, and its mechanism may be related to the down-regulation of the transcription and expression levels of connective tissue growth factor genes.
