目的 利用基因工程包装细胞进行基因转导和药物治疗相结合来治疗脑胶质瘤的可行性。方法 基因治疗。结果 实验组C_6B_PBAG+psi2STK由于psi2STK细胞的存在,其对GCV的敏感性比对照组C_6B_PBAG+psi2明显增强。结论 通过体外实验证实了逆转录病毒载体转导的HSV—TK基因,在培养的C_6胶质瘤细胞中,提高了其对药物GCV的敏感性,为下一步的动物模型及最终应用到临床打下了实验基础。
Objective To use genetic packaging cells for gene transduction and drug therapy to treat brain gliomas. Methods Gene therapy. Results In the experimental group, C_6B_PBAG+psi2STK was significantly more sensitive to GCV than C_6B_PBAG+psi2 in the presence of psi2STK cells. Conclusion The in vitro experiments confirmed that the HSV-TK gene transduced by retroviral vectors improves the sensitivity to GCV of drugs in cultured C_6 glioma cells, laying the foundation for the next animal model and eventually applying it to the clinical setting.
