目的欲建立一套完整的质量检测技术对遗传工程小鼠质量进行控制,以保证作为后续医药或其他生命科学动物实验研究的科学性和严谨性。方法在遗传、微生物、体内外寄生虫、病理、研发及繁育设施环境、饲料营养等常规质量检测的基础上,开展用PCR、Southern-blot检测目的基因;用RT-PCR、ELISA、SDS-PAGE、Western-blot等检测基因表达;用血液常规、血液生化及各器官形态、功能等检测来描述机体表征这三个层次综合描述遗传工程小鼠的特征。结果与结论将建立的技术方法应用于转基因小鼠、基因突变鼠、基因缺失鼠的检测,验证所建立的方法用于遗传工程小鼠质量控制的可行性。
Objective To establish a complete set of quality testing techniques to control the quality of genetically engineered mice so as to ensure scientific and rigorous experimental research as follow-up medicine or other life science animals. Methods The target genes were detected by PCR and Southern-blot on the basis of conventional quality tests such as genetic, microbe, ectoparasites in vitro and in vivo, pathology, environment of R & D and breeding facilities and feed nutrition. , Western-blot and other test gene expression; with blood routine, blood biochemical and organ morphology, function and other tests to describe the three levels of body characterization of a comprehensive description of the characteristics of genetically engineered mice. RESULTS AND CONCLUSION The established method was applied to the detection of transgenic mice, gene mutation mice and gene deletion mice and the feasibility of the established method was verified for the quality control of genetically engineered mice.
