目的建立人精子间期核三色荧光原位杂交(tripl-color fluorescence in situhybr idizat ion)的实验方法。方法采用三色荧光原位杂交技术对10例和正常精子标本进行间期核的原位杂交,并统计精子间期核13、16、18、21、22、X、Y染色体的杂交信号数量。结果在显微镜下可见精子头部有杂交探针的信号,间期核背景经DAPI复染显示蓝色。结论本法具有荧光杂交信号直观易分辨、短时间内能大量分析精子数量和实验操作相对简便、结果准确可信等优点。
Objective To establish an experimental method of human sperm interchromatic triple in situ hybridization (tripl-color fluorescence in situ hybridization). Methods Tricolor fluorescence in situ hybridization (ISSR) was used to in situ hybridize the interphase nuclei of 10 spermatozoa with normal spermatozoa. The number of hybridization signals of 13, 16, 18, 21, 22, X and Y chromosomes was also calculated. Results Under the microscope, there was a signal of hybridization probe in the head of spermatozoa, and the interphase nucleus background was blue by DAPI staining. Conclusion This method has the advantages of direct and easy discrimination of fluorescent hybridization signals, large amount of sperm analysis in a short time, relatively simple and convenient experimental operation, accurate and credible results.
