作者研究了18例慢性B 细胞性白血病患者[其中16例慢性淋巴细胞性白血病(CLL),2例慢性淋巴肉瘤细胞性白血病(LSCL)]的新分离的恶性B 细胞被NK细胞裂解的敏感性。靶细胞为病人外周血经密度梯度沉降法分离、羰基铁吞噬吸附法除去单核细胞、再经AET-E 玫瑰花密度梯度离心法除去T 细胞,所得B 细胞经间接荧光法检测纯度在95%以上。效应细胞为正常人外周血经密度梯度离心法分离及粘附法去除单核细胞后的淋巴样细胞。用~(51)Cr 释放法测定裂解效应。结果发现有14例病人的恶性B 细胞可被正常人NK 细胞裂解,效应细胞经干扰素作用后,裂解作用可明显提高,
The authors studied the sensitivity of newly isolated malignant B cells from 18 patients with chronic B-cell leukemia [16 of them chronic lymphocytic leukemia (CLL) and 2 of chronic lymphocytic sarcoma leukemia (LSCL)] to lysis by NK cells . The target cells were separated from the peripheral blood of patients by density gradient sedimentation, and mononuclear cells were removed by carbonyl iron phagocytosis and then T cells were removed by AET-E Rose Density Gradient centrifugation. The purity of the obtained B cells was 95% the above. The effector cells were isolated from normal human peripheral blood by density gradient centrifugation and the lymphoid cells were removed after mononuclear cell adhesion. Cleavage effect was determined by ~ (51) Cr release method. The results found that 14 patients with malignant B cells can be normal NK cells lysis, effector cells by interferon, the role of lysis can be significantly improved,
