目的:探讨RNA干扰沉默果蝇zeste基因增强子同源物2(enhancer of zeste homolog 2,EZH2)基因对卵巢癌细胞株A2780的Drosha基因表达的影响。方法:获得稳定表达EZH2短发夹状RNA(short hairpin RNA,shRNA)的人卵巢癌细胞株A2780-shEZH2,实时定量PCR法(qRT-PCR)和免疫印记法(Western blot)检测转染株的Drosha基因表达。结果:以未转染组A2780细胞Drosha的表达水平为100%,EZH2-shRNA转染组A2780-shEZH2细胞Drosha的mRNA及蛋白水平分别上升(53.02±4.32)%和(51.26±1.35)%,与未转染组比较差异有统计学意义(P<0.001)。结论:沉默EZH2基因可以上调Drosha基因的表达。
Objective: To investigate the effect of RNA interference on the Drosha gene expression in ovarian cancer cell line A2780 induced by zebrafish zeste homolog 2 (EZH2) gene. Methods: The human ovarian cancer cell line A2780-shEZH2 stably expressing EZH2 short hairpin RNA (shRNA) was obtained. The expression of EZH2 was detected by qRT-PCR and Western blot Drosha gene expression. Results: The Drosha expression level of A2780 cells in untransfected group was 100%, and the mRNA and protein levels of Drosha in A2780-shEZH2 cells transfected with EZH2-shRNA increased by (53.02 ± 4.32)% and (51.26 ± 1.35)%, respectively The difference between the untransfected group was statistically significant (P <0.001). Conclusion: Silencing EZH2 gene can up-regulate Drosha gene expression.
