目的:建立同时测定3种近缘植物仙鹤草、委陵菜和翻白草中芦丁、金丝桃苷、木犀草苷、槲皮素、芹菜素、山柰酚6种黄酮类化合物含量的UHPLC法,并比较3种药用植物鲜品和干品中6种黄酮类化合物的含量。方法:样品用70%乙醇超声提取0.5 h,采用Acquity HSS T3柱(100 mm×3.0 mm,1.8μm),以0.3%冰乙酸溶液(A)-乙腈(B)为流动相,梯度洗脱,流速为0.4 m L/min,检测波长为360 nm,柱温为35℃。结果:3种植物干品中,芦丁和金丝桃苷的含量普遍较高,山柰酚含量极低;委陵菜中芹菜素含量达到0.028 mg/g,翻白草和仙鹤草中未检测出芹菜素;仙鹤草中木犀草苷含量较高,而翻白草和委陵菜中木犀草苷含量较低。委陵菜和翻白草的鲜品中6种黄酮总含量高于干品,而仙鹤草反之。结论:3种近缘植物中委陵菜和翻白草的鲜品与干品黄酮含量变化趋势相同,从化学分类角度证实两者更为近缘。该方法简单、快速且高效,应用该方法分析3种近缘植物中黄酮类成分的含量差异,为临床合理用药提供了科学依据。
OBJECTIVE: To establish a method for the simultaneous determination of six flavonoid compounds including rutin, hyperoside, luteolin, quercetin, apigenin and kaempferol in three species of Agrimony, UHPLC method, and compared the content of 6 kinds of flavonoids in 3 kinds of fresh and dried medicinal plants. Methods: The samples were extracted with 70% ethanol for 0.5 h. The samples were eluted with 0.3% glacial acetic acid (A) - acetonitrile (B) using the Acquity HSS T3 column (100 mm × 3.0 mm, 1.8 μm) The flow rate was 0.4 m L / min, the detection wavelength was 360 nm and the column temperature was 35 ℃. Results: The contents of rutin and hyperoside in the three kinds of plant dried products were generally high, and the content of kaempferol was very low. The content of apigenin in Potentilla anserine reached 0.028 mg / g, Detected apigenin; agrimony grass luteolin content is higher, while the white grass and Potentilla Luteolin content is low. The total content of six flavonoids in the fresh products of Potentilla anserine and Turfgrass was higher than that of dried products, whereas the agrimony was the other way around. CONCLUSION: The changes of flavonoids content of fresh and dried flavescens in Potentilla anserine and Turpan blossoms in the three species of relatives are the same, and they are more close to each other from the perspective of chemical classification. The method is simple, rapid and efficient. The application of the method to analyze the content of flavonoids in three species of relative plants provides a scientific basis for clinical rational drug use.
