目的:筛选分光光度法测定茯苓中多糖和总三萜类活性成分的最优条件,以便快速准确地进行茯苓饮片的质量评价。方法:采用苯酚-浓硫酸比色法测定茯苓多糖的含量,分别从沸水浴时间、苯酚和浓硫酸用量等方面考察最优条件;采用香草醛-冰乙酸、高氯酸比色法测定茯苓三萜类成分的含量,分别从5%香草醛-冰乙酸溶液用量、高氯酸用量、水浴温度、水浴时间等方面考察最优条件;同时进行精密度、重复性、稳定性、加样回收率等方面的考察。结果:茯苓多糖测定的优化条件为苯酚1.0mL、浓硫酸5.0mL、沸水浴20min,糖含量在10~100μg范围与其峰面积呈良好的线性关系,回归方程为Y=0.008 4X-0.000 9(R2=0.999 2);总三萜类成分优化条件为5%香草醛-冰乙酸0.2mL、高氯酸1.0mL、70℃水浴20min,三萜类成分10~70μg范围内与其峰面积呈良好的线性关系,回归方程为Y=0.0114X-0.006 7(R2=0.999 9)。结论:此法操作简便,测定结果准确可靠,可用于茯苓多糖和总三萜类含量的分析。
OBJECTIVE: To screen the optimal conditions for the determination of polysaccharides and total triterpenoids in Poria cocos by spectrophotometry in order to evaluate the quality of Poria Pieces quickly and accurately. Methods: The content of polysaccharide was determined by phenol-concentrated sulfuric acid colorimetric method. The optimal conditions were investigated respectively from the time of boiling water bath, the amount of phenol and concentrated sulfuric acid. The content of polysaccharides was determined by the method of vanillin-glacial acetic acid and perchloric acid The optimum conditions were determined from the content of 5% vanillin-glacial acetic acid solution, the amount of perchloric acid, the temperature of water bath and the time of water bath. The precision, repeatability, stability and sample recovery And other aspects of the inspection. Results: The optimal conditions for the determination of tuckahoe polysaccharide were phenol 1.0mL, concentrated sulfuric acid 5.0mL, boiling water 20min, sugar content in the range of 10 ~ 100μg and its peak area showed a good linear relationship, the regression equation was Y = 0.008 4X-0.000 9 (R2 = 0.999 2). The optimal conditions for total triterpenoids were 0.2% vanillin-glacial acetic acid, 1.0 mL perchloric acid and 20 min water bath at 70 ℃. The triterpenoids showed good linearity with their peak area in the range of 10-70 μg The regression equation is Y = 0.0114X-0.006 7 (R2 = 0.999 9). Conclusion: The method is simple, accurate and reliable, and can be used for the analysis of the content of tuckahoe and total triterpenoids.
