探讨群落差异下油松居群适应性和变异特征对油松资源管理和利用具有重要意义.采用ISSR分子标记方法检测我国特有树种油松居群的遗传变异,分析3个居群间的遗传分化水平,探讨影响群体遗传分化的主要因素和机制.结果表明,13个ISSR引物共检测到185个可重复的位点,其中多态位点168个,总多态位点百分率(P)为90.81%,平均多态位点百分率为66.38%;3个油松居群间的遗传分化系数Gst平均为0.178 1,表明群体间的遗传分化水平较高,遗传变异82.19%存在于群体内,居群间的基因流(Nm=2.3081)相对异交风媒植物(Nm=5.380)较小;3个植物群落油松居群的Nei’s遗传多样性指数大小顺序为:油松+核桃楸+白桦混交林(YHB)>油松纯林(YY)>油松+华北落叶松+白桦混交林(YLB);居群间的遗传相似度平均为0.902 7;油松居群的遗传多样性与枝下高和辛普森指数呈显著正相关性;油松居群遗传变异与居群的群落特征关系密切.不同群落类型生境影响油松遗传多样性水平.
It is of great significance to explore the adaptability and variation characteristics of Pinus tabulaeformis populations to community management and utilization of Pinus tabulaeformis populations with community differences.The ISSR markers were used to detect the genetic variation of Pinus tabulaeformis populations in China and the genetic differentiation among the three populations was analyzed. The main factors and mechanisms influencing population genetic differentiation were explored.The results showed that a total of 185 repeatable loci were detected by 13 ISSR primers, of which 168 polymorphic loci, the percentage of total polymorphic loci (P) was 90.81% The average percentage of polymorphic loci was 66.38%. The genetic differentiation coefficient Gst of three populations was 0.178 1, indicating that the genetic differentiation among populations was high, and 82.19% of the genetic variation existed in the population. (Nm = 2.3081) was smaller than that of the other plants (Nm = 5.380). Nei’s genetic diversity index of Pinus tabulaeformis population in three plant communities was: Pinus tabulaeformis + Betula platyphylla + Betula platyphylla forest (YHB) (YY)> Pinus tabulaeformis + Larix principis-rupprechtii + Betula platyphylla (YLB). The average genetic similarity among populations was 0.902 7. The genetic diversity of Pinus tabulaeformis population was significantly correlated with the branch height and Simpson index Positive correlation; Pinus tabulaeformis populations genetic variation and population groups Features close. Different Communities habitats affect the level of genetic diversity of Chinese Pine.
