目的:探讨人终丝匀浆上清液体外诱导骨髓间充质干细胞分化为神经样细胞的可行性。方法:将体外培养的人骨髓间充质干细胞分为实验组和对照组。实验组加入无血清培养基及人终丝匀浆上清液诱导,对照组只加培养基,>72 h进行免疫细胞化学鉴定。结果:诱导后有17%的细胞发生典型的形态学变化。神经丝蛋白阳性率15.9%,神经元特异性烯醇化酶阳性率13.7%,神经胶质纤维酸性蛋白阳性率18.3%。结论:终丝中含有神经生长必备的细胞因子,并能诱导骨髓间充质干细胞分化为神经样细胞。
Objective: To investigate the feasibility of human marrow homogenate supernatant inducing differentiation of bone marrow mesenchymal stem cells into neuron-like cells in vitro. Methods: Human bone marrow mesenchymal stem cells cultured in vitro were divided into experimental group and control group. The experimental group was added with serum-free medium and human silk fibroin supernatant to induce, the control group only with medium,> 72 h immunocytochemistry. Results: 17% of cells underwent typical morphological changes after induction. Neurofilament positive rate of 15.9%, neuron specific enolase positive rate of 13.7%, glial fibrillary acidic protein positive rate of 18.3%. Conclusion: The terminal silk contains the necessary cytokines for nerve growth and induces the differentiation of bone marrow mesenchymal stem cells into neuron-like cells.
