目的观察五紫三黄方对醋酸泼尼松片干预治疗的哮喘模型小鼠下丘脑—垂体—肾上腺(HPA)轴功能的影响。方法将120只BALB/C小鼠随机分为空白组、模型组、醋酸泼尼松片组和五紫三黄方低、中、高剂量组,每组20只。空白组小鼠用生理盐水腹腔注射和雾化,其余5组小鼠于第1、7、14天给予腹腔注射10μg卵蛋白+2mg氢氧化铝凝胶;第21天开始用1%卵蛋白雾化激发,每天1次,连续7天,其后每隔3天雾化1次,共雾化18次。每次雾化前醋酸泼尼松片组和五紫三黄方低、中、高剂量组小鼠均给予3.9 mg/kg醋酸泼尼松灌胃,第42天开始按照每两天醋酸泼尼松0.325 mg/kg速度撤减激素用量,于第63天全部撤离;同时从第35天开始五紫三黄方低、中、高剂量组分别给予五紫三黄方配方颗粒1.994g/kg、3.988 g/kg、7.976 g/kg灌胃,模型组和空白组给予等体积的生理盐水;第64天采用放射免疫法测定各组小鼠血浆促肾上腺皮质素释放激素(CRH)、促肾上腺皮质激素(ACTH)和皮质酮(COR)含量。结果模型组与空白组小鼠血浆COR、CRH和ACTH含量比较差异无统计学意义(P>0.05);与模型组比较,醋酸泼尼松片组小鼠以上指标均降低(P<0.05或P<0.01);与醋酸泼尼松片组比较,五紫三黄方低、中、高剂量组以上指标均有升高(P<0.05或P<0.01),且五紫三黄方高剂量组的改善程度优于五紫三黄方低、中剂量组(P<0.05或P<0.01)。结论五紫三黄方能够改善醋酸泼尼松片干预治疗哮喘小鼠导致的HPA轴功能受损,且大剂量使用效果更佳。

Results There was no significant difference in the plasma levels of COR, CRH and ACTH between the model group and the blank group (P> 0.05). Compared with the model group, the above indexes in the prednisone acetate group mice were lower (P <0.05 or P <0.01). Compared with the prednisone acetate group, the above five indicators of low, medium and high doses of WZSYG were significantly increased (P <0.05 or P <0.01) The degree of superiority was better than that of Wanshuanghuanghuang recipe (P <0.05 or P <0.01). Conclusion Wenshen Huanghuang decoction can improve the impairment of HPA axis caused by the intervention of prednisone acetate tablets in asthmatic mice, and the effect of high dose is better.
