目的观察法舒地尔对家兔脊髓缺血再灌注损伤后细胞间粘附分子-1(ICAM-1)表达水平的影响。方法家兔42只,随机分为假手术组、模型组、法舒地尔组,采用腹主动脉夹闭法制作缺血再灌注模型,法舒地尔组每日腹腔注射法舒地尔,模型组注射等量生理盐水,假手术组不予干预。手术后采用Jacobs法对家兔后肢功能评级,肌电图检测后肢运动诱发电位。免疫组化法观察脊髓缺血区ICAM-1的表达水平。结果模型组ICAM-1表达明显,后肢瘫痪严重,法舒地尔能显著抑制ICAM-1的表达,瘫痪程度较轻,后肢运动诱发电位潜伏期缩短。结论法舒地尔可能通过抑制Rho激酶活性,进而抑制ICAM-1的表达,减轻脊髓缺血再灌注损伤的炎性反应,达到保护作用。
Objective To observe the effect of fasudil on the expression of intercellular adhesion molecule-1 (ICAM-1) after spinal cord ischemia-reperfusion injury in rabbits. Methods Forty-two rabbits were randomly divided into sham-operation group, model group and fasudil group. The models of ischemia-reperfusion were established by abdominal aorta clamping. Fasudil was given intraperitoneally to fasudil group, Model group was injected with normal saline, sham-operated group without intervention. After the operation, the hindlimb function of rabbits was graded by Jacobs method and the electromyogram was used to detect the evoked potentials of hind limbs. Immunohistochemistry was used to observe the expression of ICAM-1 in spinal cord ischemic area. Results The expression of ICAM-1 in the model group was significant. The paralyzed hindlimb was serious. Fasudil could significantly inhibit the expression of ICAM-1. The degree of paralysis was mild and the latent period of motor evoked potentials of the hindlimb was shortened. Conclusion fasudil may protect the spinal cord ischemia-reperfusion injury by inhibiting the activity of Rho kinase, thereby inhibiting the expression of ICAM-1.
