目的:观察丹酚酸B对STZ联合高脂饲养ApoE-/-小鼠主动脉Bcl-2和Bax蛋白表达的影响。方法:8周龄ApoE-/-小鼠,除空白对照组外,STZ联合高脂饲养12周后,随机分为4组:①模型组;②洛伐他汀组;③丹酚酸B高剂量组;④丹酚酸B中剂量组。药物连续灌胃8周后,采用免疫组化方法检测各组大鼠主动脉Bcl-2和Bax蛋白表达量以及二者比值。结果:与空白对照组比较,模型组Bcl-2和Bax蛋白表达量明显增加,以后者更为显著,二者比值失调;丹酚酸B高剂量组Bcl-2蛋白表达量增加,同时Bax蛋白表达降低,二者比值恢复。结论:丹酚酸B能上调Bcl-2蛋白表达以及下调Bax蛋白表达,并恢复二者比值,可能是丹酚酸B防治糖尿病动脉粥样硬化的作用机制之一。
AIM: To observe the effect of salvianolic acid B on the protein expressions of Bcl-2 and Bax in the aorta of STZ combined with high-fat diet ApoE - / - mice. Methods: ApoE - / - mice of 8 weeks old were randomly divided into 4 groups: ① model group, ② lovastatin group, ③ high dose of salvianolic acid B Group; ④ salvianolic acid B medium dose group. After 8 weeks of continuous gavage, the protein expression of Bcl-2 and Bax in the aorta of rats in each group were detected by immunohistochemistry and their ratios were compared. Results: Compared with the blank control group, the expression of Bcl-2 and Bax in the model group increased significantly, the later was more significant, the ratio of the two was imbalance; Bcl-2 protein expression increased in the high-dose salvianolic acid B group, Decreased expression, the ratio between the two restored. Conclusion: Salvianolic acid B can up-regulate Bcl-2 protein expression and down-regulate Bax protein expression, and restore the ratio between them. It may be one of the mechanisms of salvianolic acid B in prevention and treatment of diabetic atherosclerosis.
