目的探讨过氧化物酶增殖体激活受体-δ(PPAR-δ)基因多态性与淮海地区汉族人群2型糖尿病(T2DM)及代谢指标的相关性,以期进一步了解T2DM发病的遗传学机制。方法采用聚合酶链反应-限制性片断长度多态性(PCR-RFLP)方法对200例T2DM患者(病例组)和200例健康人群(对照组)进行基因多态性检测。同时测定两组人群的身高、体重、腰围、臀围、收缩压(SBP)、舒张压(DBP),空腹血糖(FPG)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)等临床指标,计算体质指数(BMI)和腰臀比。结果 PPAR-δ-87T/C位点基因型和等位基因频率在病例组和对照组间差异均无统计学意义(P>0.05)。此位点不同基因型T2DM患者的体重和BMI差异有统计学意义(P<0.05);两两比较结果显示,CC型的体重([76.75±11.10)kg]显著高于TT型([68.43±13.10)kg],CC型的BMI([27.99±3.62)kg/m2]显著高于TT和TC型[分别为(25.10±3.71)、(25.69±3.40)kg/m2],差异有统计学意义(P<0.05,P<0.01)。其他指标如年龄、身高、腰围、臀围、腰臀比、SBP、DBP、FPG、TC、TG、HDL-C、LDL-C在3组基因型患者中差异均无统计学意义(P>0.05)。结论 PPAR-δ-87T/C多态性可能与中国淮海地区汉族人群2型糖尿病的发病无关,其多态性可能与2型糖尿病患者的肥胖发生有一定相关性。
Objective To investigate the relationship between peroxisome proliferator activated receptor-δ (PPAR-δ) gene polymorphism and type 2 diabetes mellitus (T2DM) and metabolic parameters in the Han population in Huaihai, China. In order to further understand the genetic mechanism of T2DM. Methods 200 cases of T2DM patients (case group) and 200 healthy people (control group) were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. The changes of body weight, waist circumference, hip circumference, SBP, DBP, FPG, TC, TG, HDL Cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C) and other clinical indicators, body mass index (BMI) and waist-hip ratio were calculated. Results There was no significant difference in genotype and allele frequency between PPAR-δ-87T / C locus and control group (P> 0.05). The weight and BMI of T2DM patients with different genotypes at this site were significantly different (P <0.05). The results of pairwise comparison showed that the body weight of CC type (76.75 ± 11.10 kg) was significantly higher than that of TT genotype (68.43 ± 13.10 kg and CC type BMI were significantly higher than those of TT and TC [(25.10 ± 3.71) and (25.69 ± 3.40) kg / m 2, respectively], with significant difference (P <0.05, P <0.01). There were no significant differences in age, height, waist circumference, hip circumference, waist-hip ratio, SBP, DBP, FPG, TC, TG, HDL-C and LDL-C among the three genotypes ). Conclusion Polymorphisms of PPAR-δ-87T / C may not be related to the incidence of type 2 diabetes in Han population in Huaihai, China. The polymorphism of PPAR-δ-87T / C may be related to the occurrence of obesity in type 2 diabetes mellitus.
