目的观察BMP-2基因在静脉移植模型中的动态表达及与内膜增生的关系。方法建立兔自体静脉移植动物模型,分别于术后3、7、14、28d取材。Verhoeff染色观察各时间点内膜厚度及管壁厚度;Western blot检测BMP-2蛋白的变化,免疫组化(SABC)法检测BMP-2、增殖细胞核抗原(PCNA)的表达。结果术后7d内膜明显增厚,14d内膜增厚达到高峰(P<0.01)。BMP-2蛋白在正常对照血管可见表达,术后3d内无表达,术后第七天开始表达,第十四天明显增高,28d达到高峰(P<0.01)。PC-NA的表达于术后7~14d达到高峰。结论 BMP-2基因的表达可能是抑制移植静脉内膜增生的环节之一,可能成为治疗移植血管再狭窄的目的基因。
Objective To observe the dynamic expression of BMP-2 gene in vein graft model and its relationship with intimal hyperplasia. Methods Rabbit autologous vein graft animal model was established, respectively 3,7,14,28 d after surgery. Verhoeff staining was used to observe the intima thickness and wall thickness at each time point. The protein expression of BMP-2 was detected by Western blot. The expression of BMP-2 and PCNA was detected by immunohistochemistry (SABC). Results The intima was thickened 7 days after operation and the intima thickening reached the peak on 14th day (P <0.01). The expression of BMP-2 protein in the normal control group was not seen in the 3d control group. The expression of BMP-2 protein began on the seventh day after the operation, and increased significantly on the fourteenth day and peaked on the 28th day (P <0.01). The expression of PC-NA peaked at 7 ~ 14 days after operation. Conclusion The expression of BMP-2 gene may be one of the mechanisms that inhibit intimal hyperplasia of graft vein and may be the target gene for the treatment of vascular restenosis.
