目的探讨δ-catenin在肾透明细胞癌(Clear cell renal cell carcinoma,cc RCC)组织中的表达情况,并分析其临床意义。方法采用免疫组化SP法和Western blot检测98例cc RCC组织及40例癌旁组织中δ-catenin蛋白的表达;应用Real-time PCR检测20例cc RCC及癌旁新鲜组织中δ-catenin m RNA的表达;分析δ-catenin蛋白表达与临床病理参数及预后的关系。结果δ-catenin m RNA和蛋白在cc RCC中表达水平显著高于癌旁正常组织(<0.05)。δ-catenin蛋白的阳性表达与cc RCC患者性别、淋巴结转移及血行转移相关(<0.05)。δ-catenin蛋白高表达的患者预后明显差于低表达的患者(<0.05)。COX多因素回归分析显示,δ-catenin蛋白表达水平是影响cc RCC患者预后的独立危险因素。结论δ-catenin在cc RCC发生发展中发挥重要作用,可作为cc RCC预后判断的生物标志物。
Objective To investigate the expression of δ-catenin in clear cell renal cell carcinoma (CCCC) and to analyze its clinical significance. Methods The expression of δ-catenin protein in 98 cases of cc RCC tissues and 40 cases of adjacent tissues was detected by immunohistochemical SP method and Western blot. The expression of δ-catenin in 20 cases of cc RCC and adjacent tissues was detected by Real-time PCR RNA expression; analysis of δ-catenin protein expression and clinicopathological parameters and prognosis. Results The expression of δ-catenin m RNA and protein in cc RCC was significantly higher than that in adjacent normal tissues (<0.05). The positive expression of δ-catenin was associated with gender, lymph node metastasis and hematogenous metastasis in patients with cc RCC (<0.05). Patients with high expression of δ-catenin had significantly worse prognosis than those with low expression (<0.05). COX multivariate regression analysis showed that the expression of δ-catenin protein was an independent risk factor for the prognosis of patients with cc RCC. Conclusion δ-catenin plays an important role in the development of cc RCC and may be used as a biomarker for the prognosis of cc RCC.
