目的:研究肾上腺髓质素(AM)对内皮素1(ET-1)、血管紧张素(Ang)促肺动脉平滑肌细胞增殖的影响。方法:体外培养兔肺动脉平滑肌细胞,采用3H-TdR掺入法观察不同剂量AM对ET-1,Ang促肺动脉平滑肌细胞蛋白质合成的影响。结果:1×10-9mol/L的ET-1,1×10-8mol/L的Ang明显增加培养的平滑肌细胞3H-TdR掺入量。1×10-7~2×10-8mol/L浓度的AM明显抑制1×10-9mol/L的ET-1引起的平滑肌细胞3H-TdR掺入量的增加;1×10-7~5×10-8mol/L浓度的AM明显抑制1×10-8mol/L的Ang引起的平滑肌细胞3H-TdR掺入量的增加。结论:AM能剂量依赖地抑制ET-1,Ang促肺动脉平滑肌增生,可能在肺血管病理变化中起一定作用。
Objective: To investigate the effect of adrenomedullin (AM) on the proliferation of pulmonary artery smooth muscle cells induced by endothelin-1 (ET-1) and angiotensin (Ang). Methods: Rabbit pulmonary artery smooth muscle cells were cultured in vitro. 3H-TdR incorporation was used to observe the effect of different doses of AM on protein synthesis of pulmonary artery smooth muscle cells induced by ET-1 and Ang. Results: 1 × 10-9mol / L ET-1, 1 × 10-8mol / L Ang-3 significantly increased 3H-TdR incorporation in cultured smooth muscle cells. 1 × 10-7 ~ 2 × 10-8mol / L AM significantly inhibited 1 × 10-9mol / L ET-1-induced 3H-TdR incorporation of smooth muscle cells increased; 1 × 10-7 ~ 5 × The concentration of 10-8mol / L AM significantly inhibited 1 × 10-8mol / L of Ang-induced 3H-TdR incorporation of smooth muscle cells. Conclusion: AM can inhibit the proliferation of pulmonary artery smooth muscle cells in a dose-dependent manner and may play a role in the pathological changes of pulmonary vessels.
