目的建立测定家兔血浆中银(Ag)浓度的电感耦合等离子体质谱(ICP-MS)方法,用以研究家兔阴道给予纳米银凝胶后其在体内的吸收特征。方法样品经高压消解,以铑(Rh)为内标,采用ICP-MS法测定血浆中Ag的浓度。结果以水为替代基质建立标准曲线,Ag的线性为1.25~40.00μg·L-1(r=0.997 9),日内和日间精密度RSD≤4.7%。以水为替代基质时Ag的平均提取回收率在96.6%~98.6%内,以空白血浆为基质时Ag的提取回收率为94.8%。以水和空白血浆为基质的基质效应均符合有关规定。家兔血浆中Ag的本底水平较低。家兔多次阴道给予纳米银凝胶后,部分家兔血浆中银的浓度相比于本底水平有一定的增加。结论本方法可用于研究纳米银医用材料在家兔体内的吸收特征。
Objective To establish an ICP-MS method for the determination of silver (Ag) in rabbit plasma, and to study its in vivo absorption characteristics after vaginal administration of nanosilver gel. Methods Samples were subjected to high pressure digestion and rhodium (Rh) was used as an internal standard. The concentration of Ag in plasma was determined by ICP-MS. Results The standard curve was established with water as the substitute matrix. The linearity of Ag was 1.25-40.00 μg · L-1 (r = 0.997 9). The intra-and inter-day precision RSD was less than or equal to 4.7%. The average extraction recovery rate of Ag in water was 96.6% ~ 98.6%. The blank recovery of Ag was 94.8%. The matrix effects with water and blank plasma were in accordance with the relevant regulations. The background level of Ag in rabbits plasma is lower. After repeated vaginal administration of nanosilver gel in rabbits, the plasma concentration of silver in some rabbits increased to some extent compared with the background level. Conclusion This method can be used to study the absorption characteristics of nano-silver medical materials in rabbits.
