目的研究淋巴滞留性脑病对自主神经系统的影响。方法采用阻断颈部淋巴引流的方法将Wistar大鼠80只制作成淋巴滞留性脑病(LE)模型,并随机分为假手术组(SHAM组)和淋巴滞留性脑病组(LE组)各40只,每组又分为术后1、7、14、21 d 4个子组。采集肢体Ⅱ导联心电信号,进行心率变异性(HRV)分析,观察延髓头端腹外侧(RVLM)区脑组织形态学变化,用Western blot方法检测RVLM区eNOS蛋白的表达。结果与SHAM组相比,LE组心率变异性增大、心率减慢(P<0.05),迷走神经调节增强,RVLM区出现神经细胞变性坏死,毛细血管通透性增加,脑组织水肿,超微结构可见神经细胞凋亡、髓鞘破裂,eNOS蛋白表达下调。结论 LE造成大鼠脑组织结构与自主神经调节功能损伤,损伤的机制可能部分与下调eNOS蛋白表达有关。
Objective To study the effects of lymphostatic encephalopathy on the autonomic nervous system. Methods Eighty Wistar rats were divided into sham-operated group (SHAM group) and lymphatic encephalopathy group (LE group) by blocking the lymphatic drainage in the neck. Only, each group is divided into 4 subgroups after 1, 7, 14, 21 days after operation. The heart electrocardiogram (ECG) signals of limbs II were collected and analyzed by heart rate variability (HRV). Morphological changes of the brain were observed in the rostral ventrolateral medulla (RVLM). The expression of eNOS protein in RVLM was detected by Western blot. Results Compared with SHAM group, the heart rate variability increased, the heart rate slowed down (P <0.05), the vagal modulation was enhanced, the degeneration and necrosis of nerve cells in RVLM area, the increase of capillary permeability, the edema of brain tissue, the ultrastructure Visible neuronal apoptosis, myelin sheath rupture, eNOS protein expression down. Conclusion The mechanism of LE-induced damage of brain tissue and autonomic nervous system may partly relate to down-regulation of eNOS protein expression.
