目的探讨老化肝脏枯否细胞(KC))吞噬功能降低和对内毒素(LPS)易感性的机制.方法体外观察不同浓度 LPS对不同月龄(6,12,18和24)大鼠肝脏 KC 肌动蛋白、肌球蛋白、波形蛋白三种细胞骨架成份的影响.结果与6月龄组比较,18和24月龄组 KC 三种骨架成份分布明显改变,含量明显降低;6和24月龄组 KC 受低浓度 LPS(10ng/ml)刺激后各细胞骨架成分明显增加,但24月龄组增加幅度显著低于6月龄组;受中浓度 LPS(100ng/ml)刺激时细胞骨架较低浓度刺激时呈降低趋势,并以24月龄组为显著;受高浓度 LPS(10ug/ml)刺激时,6和24月龄骨架成分均有明显降低,两月龄组之间无统计学差异.结论老化 KC 骨架成份含量降低及对 LPS 剂量耐受性降低可能是其吞噬功能降低及受损易感性增高的重要原因之一.
Objective To investigate the mechanism of phagocytosis and susceptibility to endotoxin (LPS) in aged liver Kupffer cell (KC) .Methods The effects of different concentrations of LPS on liver KC muscle of rats of different ages (6, 12, 18 and 24) Actin, myosin and vimentin.Results Compared with the 6-month-old group, the distribution of KC three skeletons in 18 and 24 month old groups significantly changed, After stimulated with low concentration of LPS (10ng / ml), KC significantly increased the cytoskeleton components of the cells, but the increase in 24-month-old mice was significantly lower than that in the 6-month-old mice. When stimulated by LPS (10ug / ml), the skeleton components at 6 and 24 months of age were significantly decreased, but there was no significant difference between two-month-old groups. Conclusion The decreased content of aging KC and the decrease of LPS dose tolerance may be one of the important reasons for the decreased phagocytic function and increased susceptibility to damage.
