无血清培养基中的Wistar大白鼠脾细胞在ConA的刺激下可以产生IL-2.以T淋巴母细胞的增殖为指标,作者对含FCS的培养基和无血清培养基作了比较;证明在适宜的条件下,两种方法得到的IL-2滴度在峰值及动力学等方面都相同,唯一的差别是前者所需细胞浓度为1×10~7细胞/ml,而后者则为5×10~6细胞/ml.因此,从单一细胞的角度来看,有血清培养基比无血清培养基的效果好.以往以为,PMA(Phorbol Myristic Acetate)的刺激以及细胞的期前孵育都可增强丝裂原诱导的人和大白鼠淋巴细胞产生IL-2.但作者在ConA刺激的同时使用了PMA,却发现在一般情况下PMA对IL-2的产生无增强效应;而将淋巴细胞在无血清培养基中期前孵育则可大大提高IL-2的滴度.
