为了研究巴氏蘑菇Agaricus blazei多糖对铅中毒大鼠脾脏细胞因子mRNA表达的影响,进而探索巴氏蘑菇多糖对铅中毒大鼠的免疫调节作用。选用45日龄SD大鼠,随机分为6组,每组8只,雌雄各半。分别为正常对照组、多糖组、铅中毒模型组、铅+50mg/kg·d多糖试验组、铅+100mg/kg·d多糖试验组、铅+200mg/kg·d多糖试验组。铅中毒模型组、铅+多糖试验组分别给予含0.2%醋酸铅饮水,自由饮用。饲养60d后,采集脾脏,荧光定量RT-PCR对其细胞因子IL-1β、IL-2、IL-6、TNF-α、IFN-γmRNA的相对表达量进行分析。结果表明,与正常对照组相比,IL-1β、IL-2、IL-6、TNF-α、IFN-γmRNA的表达量,多糖组升高,差异显著(P<0.05或P<0.01);铅中毒模型组下降差异极显著(P<0.01);试验组多糖浓度越高,表达量越高,铅+200mg/kg·d多糖组升高差异极显著(P<0.01)。与铅中毒模型组相比,试验组多糖浓度越高,表达量相对升高越多,铅+200mg/kg·d多糖组升高差异极显著(P<0.01)。说明巴氏蘑菇多糖对铅致大鼠免疫损伤具有保护作用。
In order to study the effect of Agaricus blazei polysaccharides on mRNA expression of cytokines in spleen of lead poisoning rats, the immunomodulatory effect of pasteurized mushroom polysaccharide on lead poisoning rats was explored. 45-day-old Sprague-Dawley rats were randomly divided into 6 groups with 8 rats in each group. They were the normal control group, the polysaccharide group, the lead poisoning model group, the lead + 50mg / kg · d polysaccharide test group, the lead + 100mg / kg · d polysaccharide test group and the lead + 200mg / kg · d polysaccharide test group respectively.
