目的 研究钙结合蛋白 D2 8k(Ca BP)在帕金森病(PD)发病机制中的作用 .方法 将 1-甲基 - 4-苯基 - 1,2 ,3,6 -四氢吡啶 (MPTP)按 30 mg·kg- 1剂量腹腔注射给 C5 7BL 小鼠 ,建立 PD鼠模型 .用反转录 PCR及原位分子杂交方法检测 PD鼠和正常鼠脑内 Ca BP m RNA的表达变化 .结果 与正常鼠对比 ,Ca BP m RNA在 PD鼠的海马、纹状体、黑质等脑区的表达水平明显下调 .结论 Ca BP可能通过对特定脑区神经元的保护作用来抵御 PD的发生
Objective To investigate the role of calcium binding protein D2 8k (Ca BP) in the pathogenesis of Parkinson’s disease (PD) .Methods 1-Methyl-4-phenyl-1,2,6,6-tetrahydropyridine The mice were injected intraperitoneally with 30 mg · kg-1 of C5 7BL to establish the model of PD mice.The expression of Ca BP m RNA in brain of PD mice and normal mice was detected by reverse transcription PCR and in situ hybridization.Results Compared with normal mice, the expression level of Ca BP m RNA in hippocampus, striatum, substantia nigra and other brain regions was significantly decreased.Conclusion Ca BP may resist the occurrence of PD through the protection of neurons in specific brain regions
