目的 探讨白细胞介素 - 6(IL- 6)、肿瘤坏死因子 α(TNFα)对绝经后骨质疏松症骨形态计量学的影响。方法 行开腹输卵管峡部结扎后切除双侧卵巢建立绝经后骨质疏松大鼠模型 ,检测骨组织培养液中 (RPMI1 6 4 0 ) IL- 6、TNFα活性含量 ,同步用 IBAS计算机全自动图象分析系统对不脱钙组织作形态计量学测量 ,并与对照组比较。结果 去势后早期 (<4w)即有骨组织中 IL- 6、TNFα升高 (均P<0 .0 5) ,并持续在较高水平。随着绝经期的延长 (4~ 2 2 w) ,骨小梁体积 (TBV)和平均厚度 (MTT)减少 ,骨吸收表面 (RS)增大 (P<0 .0 0 2 )。骨组织中 IL- 6、TNFα和骨吸收指标在绝经后 4~ 2 2 w升高的程度无显著性。结论 骨微环境中破骨细胞释放一定量的 IL- 6、TNFα可能与绝经后骨质疏松发生有关 ,其持续增高可能致骨小梁组织结构变化。
Objective To investigate the effects of interleukin-6 (IL-6) and tumor necrosis factor-α (TNFα) on bone morphometry in postmenopausal osteoporosis. Methods The bilateral ovaries were excised after oviduct was ligated to establish the model of postmenopausal osteoporosis rats. The content of IL-6 and TNFα in bone tissue culture fluid was detected, Analysis system for non-decalcified tissue morphology measurement, and compared with the control group. Results In the early post castration period (<4w), the levels of IL-6 and TNFα in the bone tissue increased (all P <0.05) and persisted at higher levels. Trabecular bone volume (TBV) and mean thickness (MTT) decreased with the increase of menopause (4 ~ 22 w) and the surface resorption (RS) increased (P <0.002). The levels of IL-6, TNFα and bone resorption in bone tissue were not significantly increased 4 ~ 2 2 w after menopause. Conclusion Osteoclasts release a certain amount of IL-6 and TNFα in bone microenvironment, which may be related to the occurrence of postmenopausal osteoporosis. The sustained increase may lead to the change of trabecular structure.
