目的:研究山核桃叶总黄酮对小鼠胚胎间充质干细胞C3H10T_(1/2)成脂分化的影响。方法:建立C3H10T_(1/2)细胞成脂分化模型,油红O染色法分析山核桃叶总黄酮对C3H10T_(1/2)细胞成脂分化的影响;GAP-PAP酶法测定C3H10T_(1/2)细胞成脂分化过程中甘油三酯含量变化;采用荧光定量PCR和Western Blot检测过氧化物酶体增殖物激活受体γ(PPARγ)、CCAAT增强子结合蛋白α(C/EBPα)以及脂肪型脂肪酸结合蛋白4(FABP4)的mRNA和蛋白表达水平。结果:山核桃叶总黄酮能抑制C3H10T_(1/2)细胞成脂分化,20μg/m L山核桃叶总黄酮能显著减少甘油三酯的堆积,降低PPARγ、C/EBPα、FABP4的mRNA和蛋白表达水平。结论:山核桃叶总黄酮对C3H10T_(1/2)细胞成脂分化具有抑制作用,其机制可能与抑制PPARγ、C/EBPα、FABP4的表达有关。
Objective: To study the effects of total flavonoids from Hickory leaves on the adipogenic differentiation of mouse embryonic mesenchymal stem cells (C3H10T_(1/2)). METHODS: The adipogenic differentiation model of C3H10T_(1/2) cells was established. Oil red O staining was used to analyze the effect of total flavonoids from Hickory leaves on the adipogenic differentiation of C3H10T_(1/2) cells. The expression level. CONCLUSION: The total flavonoids from Hickory leaves have inhibitory effects on the adipogenic differentiation of C3H10T_(1/2) cells. The mechanism may be related to the inhibition of the expression of PPARγ, C/EBPα and FABP4.
